This study was carried out to investigate the polymorphism of Tumour Necrotic Factor Receptor Super Family 1A (TNFRSF1A) gene in Nigerian indigenous naked neck chickens. Blood samples were collected from these chickens from different geographical distributions in Oyo, Osun and Ogun states in South-West, Nigeria, followed by extraction of DNA, amplification and digestion of amplicons using MSp1. Data collected were analyzed using GenAlex software (version 6). The distribution pattern of allelic variants at the TNFRSF1A locus in Nigerian indigenous naked neck chickens was assessed using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR–RFLP). The PCR–RFLP genotyping of a 561 bp fragment of TNFRSF1A revealed the predominance of the homozygous AA genotype (Allelic frequency=0.796) compared to homozygous BB genotype (Allelic frequency=0.204). Hence, the TNFRSF1AA and TNFRSF1AB alleles exhibited different gene frequencies in the chickens sampled across the three states in South-west, Nigeria. TNFRSF1A gene being an immune gene influencing a number of immune response traits, polymorphisms in the regulatory region and their linkages with the MSp1 variants would be of great significance for genotype–phenotype association studies in Nigerian indigenous naked neck chickens.
