Abstract

Seven Indian soybean varieties were studied for their in vitro response towards callogenesis and regeneration. The morphogenic response of three type of explants viz., root, cotyledon and hypocotyl of seven varieties was observed on MS basal medium fortified with different hormone combinations. Successful plant regeneration was achieved through organogenesis intervened by callus phase from the hypocotyl explants of the cultivar JS 76–205, a black seeded variety. MS medium with cytokinin BAP 4.44microM proved to be an ideal morphogenic medium for regeneration. Among all the three explants, cotyledons exhibited higher callusing efficiency than root and hypocotyl. The auxin NAA proved better than 2,4-D for callus induction. Variety JS 335 responded best as callogenesis and direct regeneration could be obtained from apical buds in the morphogenic medium combination.
